Regulation of MonocyteChemotacticProtein–1 secretion by the Two-Pore-Domain Potassium (K2P) channel TREK-1 in human alveolar epithelial cells.

We recently proposed a role for the two-pore-domain K (+) (K2P) channels TREK-1 in the regulation of cytokine release from epithelial cells of the alveolar (AEC) by demonstrating a decrease in IL-6 secretion of TREK-1 cell deficiency, but the effect of modified TREK-1 expression of other inflammatory mediators remain poorly understood.
We are now examining the role of TREK-1 in TNF-α-induced MCP-1 release from human A549 cells. We hypothesize that the TREK-1 regulate TNF-α-induced MCP-1 secretion via c-Jun N-terminal kinase (JNK) – and protein kinase-C (PKC) -dependent pathway. In contrast to IL-6 secretion, we found that the shortage of TREK-1 resulted in increased production and secretion of MCP-1, although basic MCP-1 gene expression was unchanged in TREK-1 cell deficiency.
Unlike the TREK-1 deficiency AEC, overexpression of MCP-1 has no effect on MCP-1 secretion. Phosphorylation of JNK1 / 2/3 increase in the TREK-1 cell deficiency stimulation of TNF-α, but the pharmacological inhibition of JNK1 / 2/3 decreased MCP-1 release of both control and TREK-1 cell deficiency. Similarly, pharmacological inhibition of PKC decreased MCP-1 secretion from control and TREK-1 cell deficiency, suggesting that changes in JNK and PKC signaling pathways may not be the cause for the increase of MCP-1 secretion of TREK-1 cell deficiency.
Furthermore, MCP-1 secretion from control and TREK-1 cell deficiency is independent of extracellular Ca (2+) but sensitive to inhibition of Ca (2+) intracellular reuptake mechanism. In summary, we report for the first time that the TREK-1 deficiency in human AEC resulted in increased production of MCP-1 and secretion, and this effect appears to be related to changes in JNK-, PKC or Ca (2 +) – mediated signaling pathways in TREK- 1 cell deficiency.
Urinary chemokine (CC motif) ligand 2 ( monocytes chemotactic protein – one ) as an injury marker of tubular for early detection cisplatin-induced nephrotoxicity.
Because of the difficulty in detecting response to a special segment in the kidneys, we investigate the molecular events that underlie acute kidney injury in the proximal tubules of mice with cisplatin (cis-diamminedichloroplatinum II) induced nephrotoxicity.
Microarray analysis revealed that the mRNA levels of several cytokines and chemokines, such as interleukin-1beta, chemokine (CC motif) ligand (CCL) 2, CCL20, a chemokine (CXC motif) ligand (CXCL) 1 and CXCL10 increased significantly after treatment of cisplatin in both isolated proximal tubules and the entire kidney.
Interestingly, the level of tubular CCL2 mRNA increased immediately after administration of cisplatin, while CCL2 mRNA levels throughout the first kidney decreased and then increased. The second level of CCL2 and kidney injury molecule-1 (KIM-1) around the kidney increased after the administration of cisplatin. CCL2 immunofluorescence analysis revealed changes in the proximal tubule cells first and then in the medullary interstitium. CCL2 urinary excretion significantly increased about 3-fold compared to control day after administration of cisplatin (5mg / kg), when no changes were observed plasma creatinine and blood urea nitrogen levels.
levels of urinary KIM-1 also increased 3 times after the administration of cisplatin. In addition, CCL2 urinary KIM-1 rather than increased in rats with chronic renal failure following administration of low-dose cisplatin (2mg / kg), indicating that CCL2 bladder is selective for cisplatin-induced nephrotoxicity renal impairment.
